DIFFERENT RESPONSE OF TWO METALLOTHIONEIN SUBFAMILIES EXPOSED TO CHROMIUM (VI) IN Tetrahymena thermophila

چکیده
The expression of the heavy-metal-binding proteins, metallothioneins (MTs), are induced by heavy metals such as Cd, Cu, Zn, and Hg. MTs attenuate toxic heavy metals by sequestering them and decreasing their intracellular concentrations. However, Cr(VI) inhibits MT gene transcription in higher eukaryotes. In this study, we found that Cr affect Tetrahymena thermophila proliferation, and the lethal concentration killing 50% of the cell population (LC50) of Cr (VI) and Cr (III) is 80 μM and 3 mM, respectively. Cr (VI) upregulates the expression of CuMTs (MTT2, MTT4), but shows little effect for CdMTs (MTT1, MTT3 and MTT5) transcription in the vegetative growth stage of T. thermophila. Furthermore, upregulated expression of CdMTs gene induced by Cd (II) and Cu (II) was inhibited by Cr(VI). In contrast, upregulated expression of CuMTs gene induced by Cd (II) and Cu (II) has less change under Cr(VI) induction. CdMTs and CuMTs showed different response to Cr(VI) induction. The results further showed that the transcription regulation mechanism of CdMTs and CuMTs is different in T. thermophila.

Extreme metal adapted, knockout and knockdown strains reveal a coordinated gene expression among different Tetrahymena thermophila metallothionein isoforms
Metallothioneins (MT) constitute a superfamily of small cytosolic proteins that are able to bind metal cations through numerous cysteine (Cys) residues. Like other organisms the ciliate Tetrahymena thermophila presents several MT isoforms, which have been classified into two subfamilies (Cd- and Cu-metallothioneins). The main aim of this study was to examine the specific functions and transcrip...
متن کاملResponses of endogenous proline in rice seedlings under chromium exposure
Hydroponic experiments were performed to exam the dynamic change of endogenous proline in rice seedlings exposed to potassium chromate chromium (VI) or chromium nitrate chromium (III). Although accumulation of both chromium species in rice seedlings was obvious, more chromium was detected in plant tissues of rice seedlings exposed to chromium (III) than those in chromium (VI), majority being in...
متن کاملTetrahymena Metallothioneins Fall into Two Discrete Subfamilies
BACKGROUND Metallothioneins are ubiquitous small, cysteine-rich, multifunctional proteins which can bind heavy metals. METHODOLOGY/PRINCIPAL FINDINGS We report the results of phylogenetic and gene expression analyses that include two new Tetrahymena thermophila metallothionein genes (MTT3 and MTT5). Sequence alignments of all known Tetrahymena metallothioneins have allowed us to rationalize t...
متن کاملWhole‐cell biosensors for detection of heavy metal ions in environmental samples based on metallothionein promoters from Tetrahymena thermophila
Heavy metals are among the most serious pollutants, and thus there is a need to develop sensitive and rapid biomonitoring methods for heavy metals in the environment. Critical parameters such as bioavailability, toxicity and genotoxicity cannot be tested using chemical analysis, but only can be assayed using living cells. A whole-cell biosensor uses the whole cell as a single reporter incorpora...
متن کاملEffect of citric acid on antioxidant activity of garden cress (Lepidium sativum L.) under chromium stress
Chromium-VI would impose detrimental effects morphological, physiological, and metabolic in plants. This study aimed to research the impact of citric acid (0, 2.5 and 5 mM), on different the morphological and physiological characteristics of garden cress exposed to chromium-VI stress (0, 1, and 10 mM). Results showed that some growth parameters, chlorophyll, carotenoids, and DPPH activity decre...
